Trichrome stain was originally developed by Gomori for staining tissue sections and cytological smears.1 In 1951, Wheatley modified Gomori's technique by adding fixation and dehydration steps resulting in a simple and rapid staining procedure for intestinal amoebas and flagellates.2 PRINCIPLE Wheatley Trichrome Stain contains two dyes. Martius scarlet blue trichrome: method for staining of elastica, connective tissue in general, fibrin; stains for fresh (orange-yellow), mature (red) or old (blue) fibrin. PTAH: variant of trichrome stain; demonstrates intracytoplasmic filaments in muscle and glial cells. Stain in Trichrome Stain, Gomori One-Step, Light Green for 20 minutes. Directly differentiate in Acetic Acid 0.5%, Aqueous (100121) for 2 minutes. Rinse quickly in distilled water. Dehydrate in two changes each of 95% and 100% ethyl alcohol. Clear in three changes of xylene, 10 dips each; coverslip with compatible mounting medium. Gömöri Trichrome Stain. The modified Gomori trichrome stain specifically allows the detection of ragged-red fibers (RRF), characterized by a "fiber cracking" appearance and abnormal subsarcolemmal proliferation of mitochondria resulting from a compensatory response to a mitochondrial OXPHOS defect. Place sections in Working Weigert's Iron Hematoxylin Stain for 10 minutes. Rinse sections in tap water for 5-10 minutes. Stain sections in Trichrome Stain for 15 minutes. Place sections in 1% Acetic Acid Solution for 1 minute. Staining Procedure: Place the coverslip with section in a ceramic staining rack (Thomas Scientific #8542-E40). Immerse sections in Harris Hematoxylin for 1.5 minutes. Wash with tap water until the water is clear. Immerse sections in Gomori trichrome stain for 10 minutes. 2 ml. 5.
